The Postdoctoral Research Associate will work on athree yearPlant Genome Research Project funded by the NSF (IOS 1733633). The project is an inter-institutional research project between The University of Toledo’s Department of Biological Sciences, Michigan State University’s Department of Plant Biology, The University of Minnesota'sCollegeof Biological Sciences and the University of Wisconsin-Madison's Department of Agronomy. This project will develop resources for the investigation of transcriptional genomics in corn (Zea mays or maize). The Toledo campus is taking the lead in expanding the transcription factor (TF) ORFeome (TFome) collection) and developing anti-serum for the characterization of maize transcription factors. The Postdoctoral Research Associate will use newly developed antisera to characterize the in vivobinding properties of plant TFs of interest using techniques such as ChIP-Seq and ChIP-PCR (chromatin Immunoprecipitation with high throughput sequencing or PCR. Analysis of ChIP-Seq data will require familiarity with The Postdoctoral Research Associate will also assist in expanding the current TFome collection and then employ Information technology to screen for novel TF-promoter interactions using the Yeast-1-hybrid technique (Y1H) and purse follow-up investigations. The Postdoctoral Research Associate should have strong verbal and written communication expertise and is expected to take a leading function in developing manuscripts for publication of their work in peer reviewed journals.

Job Job responsibilities:
   » Test suitcapability of recently developed antisera for Transcription Factor (TF) sensitivity, specificity, and affinity purify as necessary.
   » Employ newly developed antisera to explore TF protein-DNA interactions (PDIs) in vivo using techniques such as ChIP-Seq and ChIP-PCR (including computational analysis of results).
   » supervise the amplification, cloning, and clone validation of maize transcription factor (TF) genes by undergraduate students using recombinant DNA technology.
   » Employ existing maize TFome (~2000 existing clones) and new clones (+ ~150) to discover new regulators of plant genes by Yeast-One-hybrid (Y1H) technique and follow up validation characterization (e.g. by reporter assays)
   » Train and supervise undergraduates to assist in the work of the Postdoctoral Research Associate
   » Take a leading function in preparing manuscripts for publication located on research conducted

Education/practice/licensing:
Ph.D. degree in Molecular Biology or related field is required.

1 year practice in a molecular biology lab is required.

Six Months practice with protein overexpression in E. coli and protein purification is required.

Six Months practice with Western/Immunoblotting blots is required.

Six Months practice with Recombinant DNA technology and amplification of DNA using the Polymerase Chain reaction (PCR) is required.
Communication and other expertise:
Able to communicate effectively with other laboratory workers (including supervision and training of undergraduate students) and present his/her results at lab meetings.

Able to write up their research findings in the English language in preparation for scientific publication
Preferred Abilities
   » Preferably has practice in the analysis of large DNA and RNA sequence datasets
   » Familiarity with DNA analysis software, database searching (BLAST program), and database entry is preferred
   » Preferably has practice in propagating and harvesting plant tissues for molecular biology studies.
   » practice in supervising of other workers is preferred

Principal Investigators's Research Interest: I am a plant molecular biologist with a current interest in regulomics in maize (corn). We have created a Transcription Factor (TF) collection of most of the TFs in the maize genome and Information technology has been used to identify novel regulatory components of the phenylpropanoid pathway - a key secondary metabolic pathway in all plants governing anthocyanin, flavonoid, and lignin biosynthesis. Our current work aims in further dissecting the regulation of phenylpropanoid genes using chromatin-immunoprecipation approaches in vivo.
